Steps To Reproducestart a clip
open the mad settings
close the clip
change something like chroma upscaling
try ok or apply

Currently settings can only be changed while a madVR instance is running. This is not a bug. It's by design. I have on my to do list to make editing settings possible even when no video is running. But that is an extra feature which will need extra code. But as I said, it's on my to do list, and it will come sooner or later.
